If you are Travelling from Public transport than you need to catch train from any central railway station towards khopoli and get down to Neral.

From Pune : Take the Mumbai-Pune Expressway and exit at Khopoli and get on to the old Mumbai-Pune highway, NH4. Ride towards Chouk Village and keep on going until you reach the base of Matheran Hill. This journey takes almost 2-4 hours as per your driving capability.
